The home side were ahead inside the opening minute when Jonny May got the ball out wide and made a quick run for the try line, stretching to make sure the ball was over before he was tackled.
Wasps soon took control, though, with Christian Wade, Tom Varndell and Nathan Hughes all scoring before half time to leave the visitors ahead at the interval.
After the break, Greig Laidlaw kicked three penalties in quick succession to cut Gloucester's lead to just a point with 20 minutes to play.
However, Andy Goode added a further three points for Wasps, before Elliott Daly took over the kicking duties to extend their lead to seven and seal the win.
The result moves Wasps up to sixth in the table, while Gloucester remain ninth.
