Things went from bad to worse for the Old Lady in the 39th minute when they were reduced to 10 men, Giorgio Chiellini receiving his marching orders for a second bookable offence.
Despite being a man down, Juve were inches away from an equaliser in the 50th minute as Mario Mandzukic held off his marker and found Paul Pogba in front of goal, but the Frenchman could only shoot wide.
It was Sassuolo's first ever victory over Juventus, and they were made to work for it as the Old Lady hit them with everything they had in the second half.
Juan Cuadrado almost found the bottom corner from inside the box just after the hour mark, but Sassuolo goalkeeper Gianluca Pegolo was equal to his strike.
Hernanes tried his luck from range after exchanging passes with Pogba with four minutes remaining, and was just inches from finding the target.
It was as close as Juventus came to an equaliser as Sassuolo held on to secure a famous win.
