The hosts were almost level immediately through Chris Zebroski, whose header had to be tipped over by Ryan Clarke following a hanging corner played into the box.
Oxford looked to be heading for a half-time lead, though with seconds remaining Lee Minshull's long-range effort took a deflection off teammate Rene Howe to draw County level.
Adam Chapman came close to firing the hosts ahead 10 minutes after the restart, but his 20-yard effort ended up going inches wide of the target to leave the scores level.
Justin Edinburgh's men did take the lead after 70 minutes, however, as Ryan Burge stood over a Newport free kick before firing it into the far corner of the net with a bending effort.
County added a third with seven minutes of normal time remaining, when Zebroski took full advantage of a mix-up between Johnny Mullins and Clarke at the back.
Alfie Potter pulled one back for the U's with seconds remaining, but it wasn't enough to stop the Welsh outfit picking up all three points.
