AJ Leitch-Smith's deflected shot presented an early opportunity for Yeovil's Sam Foley, but his shot went over the bar.
Conor Hourihane's free kick from the left was then half cleared, before Reece Brown saw an effort saved.
The hosts grew into the game as the half went on, with Joe Edwards picking out Foley on the left with a superb pass before the midfielder put his strike over the crossbar.
Their good work was undone, however, when Barnsley's Hourihane scored just before half time with a strike from 30 yards, with a slight deflection leaving Yeovil keeper Chris Weale with no chance.
Edwards unleashed a volley at goal after the restart, but Adam Davies palmed the ball down and gathered it on the second attempt.
Yeovil eventually got the equaliser through the unfortunate Martin Cranie, who put a Jakub Sokolik cross into his own net.
