World number 33 Rosol started the encounter in positive form, requiring just one hour to open up a two-set lead.
The Czech's domination continued in the next set as he broke his Spanish opponent on two occasions to wrap up the win in just 82 minutes.
Rosol will now face Georgia's Teymuraz Gabashvili for a place in the fourth round of the competition.
Should Rosol win that encounter, it would represent the deepest that he has ever gone at Roland Garros.
