The 23-year-old has already spent a similar spell at Sheffield Wednesday this season, where he featured four times for the Owls.
He returned to Upton Park at the end of September, but he will now spend the next four weeks at the League One outfit.
Spence could make his debut for the MK Dons in their away fixture at Tranmere Rovers on Saturday.
