Centre-back Jose Gimenez missed the 3-0 Group A win over Russia with a back injury but is fit again and replaces Sebastian Coates in the Celeste lineup.
The Euro 2016 winners, meanwhile, make three changes from the XI which drew 1-1 with Iran in the final match of Group B.
Ricardo Pereira, Goncalo Guedes and Bernardo Silva come in, while Cedric Soares, Ricardo Quaresma and Andre Silva make way.
Defensive midfielder William Carvalho was a doubt after missing a training session on Thursday but is deemed fit enough to start for the Selecao.
Both sides are also able to call on their star players for the encounter, as Luis Suarez lines up alongside Edinson Cavani for Uruguay and Cristiano Ronaldo is up front for Portugal.
