Morten Gamst Pedersen saw an early free kick float wide for the hosts before Jacob Mellis almost made Grant Hanley pay for an error up the other end, with Paul Robinson making a one-handed save to keep the scores level.
Both sides had plenty of attempts but it was unfancied Barnsley who went ahead when on-loan Chelsea defender Mellis lashed home a Chris Dagnall flick-on.
The Tykes couldn't hold out for a half-time lead, however, as new Rover Jordan Rhodes controlled the ball with his back to goal before hooking past Ben Alnwick.
