George Bailey top scored with 70, but one to nine all reached double figures for the Aussies against the only side above them in the ODI rankings.
David Warner and Aaron Finch put on 94 for the opening wicket before the hosts lost two wickets for no runs.
The pick of the South Africa bowlers, Vurnon Philander, had Warner caught for 46 and then found the edge of Finch (35) in the same over on his way to recording figures of 4-45 from his 10 overs.
The tourists continued to take regular wickets as Shane Watson (13), Michael Clarke (11) and Mitchell Marsh (10) all fell to restrict Australia to 144-5, but Bailey was making good progress at the other end.
He came together with wicket-keeper Matthew Wade (35) for a sixth-wicket stand of 92 during which he passed 50, but Dale Steyn came back into the attack to dismiss Bailey for 70 off 75 balls.
His partnership with Wade, who was only playing due to an injury to Brad Haddin, gave lower-order hitters Glenn Maxwell and Mitchell Johnson the platform from which to bludgeon the Aussies to a big total.
After Wade became Philander's fourth and final victim, Maxwell hit five boundaries in a 19-ball effort of 29, while Johnson made 13 from eight balls as Australia finished their innings as strongly as they started it.
