The tourists, who won the toss and chose to bat, made 70-2 during the morning session as Shikhar Dhawan (17) and Murali Vijay (six) were both dismissed having made little impact at the crease.
Cheteshwar Pujara (25) was run out as India lost their third wicket of the match after lunch before Rohit Sharma (14) was caught by AB de Villiers off the bowling of Vernon Philander.
However, Virat Kohli will start the final session of the day on 84 not out along with Ajinkya Rahane, who is on 12.
